This ELISA kit applies the two steps quantitative sandwich enzyme immunoassay technique. The microtiter plate has been pre-coated with mouse MCP-1 antibody (solid phase antigen), standards or samples are added to the microtiter plate wells, and the microtiter plate undergoes incubation. Add biotin antibody, incubate, wash thoroughly, and then add avidin enzyme. Wells are thoroughly washed to remove all unbound components, and a sandwich complex of solid antigen-antibody enzyme-labeled secondary antibodies is formed on the solid surface of the microplate. Next, Substrate solution A and substrate solution B are added to each well, and it will produce blue materials under HRP catalysis. The enzyme-substrate reaction is terminated by the addition of sulphuric acid and the color change to yellow. Measured spectrophotometrically at a wavelength of 450 nm, the absorbance (OD value) is positively correlated with the concentration of mouse MCP-1 in the sample. The concentration of MCP-1 in the samples could be determined by comparing the O.D. of the samples to the standard curve.
